CD8+ T cells, also known as killer T cells (or cytotoxic T cells), are main effector cells of the adaptive immune system by attacking invasive target cells. During an infection, naive CD8+ T cells are primed by antigen-presenting cells (APCs) in secondary lymphoid organs such as lymph nodes (LN) and the spleen, enabling them to eliminate infected or malignant cells. The killing function of CD8+ T cells is mainly mediated by Fas/Fas Ligand (FasL) system and the release of granzymes and perforins. Cytotoxic T cells are important for killing cancerous or virally infected cells and play a key role in cancer immunotherapy.  Furthermore, the development of immunotherapeutic approaches that specifically target and regulate the function of distinct CD8 T-cell subsets holds great promise for precision medicine.
